CHIP-seq for NKX2-3 overexpressing tumor pericytes

This study aims to investigate genes regulated by NKX2-3 The tumor pericytes were infected with lentiviruses carrying vector or NKX2-3 overexpression plasmids. Essentially, cells were crosslinked with formaldehyde. After chromatin sonification and centrifugation, 2% of the digested chromatin was saved and the remaining chromatin was equally divided into two aliquots, one for immunoprecipitation by anti-NKX2-3 antibody (HPA047561, Sigma, Shanghai, China), and other as the negative control by adding anti-IgG. After elution from antibody/protein G magnetic beads and reversing the cross-links, the immunoprecipitated DNA was purified and purified for sequencing.
