In situ Perturb-seq is a method for pooled optical genetic screens with spatial transcriptomics readouts in the intact tissue. Here in situ Perturb-seq was applied to A375 melanoma tumors in NSG mice to investigate top T cell receptor (TCR)-specific immunomodulating hits identified in the melanoma CRISPR activation screens.
