Detailing Novel Epigenetic Events Associated with Diet-Induced Obesity in the Rat

We applied Whole Transcriptome Termini Site Sequencing (WTTS-seq) to detect differences in use of alternative polyadenylation sites between diet-induced obesity rats and control rats. Our finding clear showed that a nutritionally complete high fat diet would affect pre-translational alternative polyadenylation events on hypothalamic transcripts that both stimulate and counter regulate body weight. Overall design: Eight adult male Long-Evans rats (Harlan, IN) were divided into obesity group and control group. Control group were maintained on standard rodent chow (3.41 kcal/g, 0.51 kcal/g from fat). Obesity group received ad libitum access to a nutritionally complete high fat diet (41 kcal/g, 1.71 kcal/g from fat). Following 36 days of diet exposure, all rats were euthanized and brain tissue collected for RNA isolation. Total RNA samples derived from eight male Long-Evan rats were used to construct WTTS-seq libraries. Therefore, this submission involved a total of 8 libraries.
