Preliminary construction of the Risk Prediction Column for preeclampsia: integration of MTHFR/VDR FokI gene polymorphism and analysis of uterine artery PI

A prospective design was adopted and the study was conducted in two phases. In the first phase (October 2022 - April 2023), 73 research subjects (42 cases in the HDP group and 30 cases in the healthy control group) were included. Genotyping and Association Analysis were performed on 8 candidate single nucleotide polymorphisms (SNPS) to screen for PE-related genetic loci. In the second stage (May 2023 - July 2025), the Sample was expanded to 195 cases (101 cases in the PE group and 94 cases in the healthy control group), candidate SNPS were verified, and clinical, ultrasound and laboratory Metric were collected. Univariate and multivariate Logistic Regression were used to screen the Independence Prediction Factor of PE, and a Column line graph model was constructed. The efficacy of the model was evaluated through Bootstrap resampling, Regression (ROC) curve, Regression curve and decision curve analysis (DCA). Meanwhile, relevant serum markers were detected, and the molecular mechanism by which MTHFR gene polymorphism affects the Risk of PE was initially explored through mediation analysis.
