基于特色的由正常肺上皮细胞到肺腺癌到肺鳞癌恶性转化的小鼠模型, 采集时序性的腺鳞癌小鼠模型临界状态的蛋白质组、磷酸化蛋白质组数据集,课题一已为课题二提供了肺腺鳞癌转分化小鼠模型(基因型KrasG12D/Lkb1-/-)的多个时间点共计17只小鼠27份肺组织样本,课题二选择其中具有3只以上生物学重复的共计5个重要时间节点共计15份小鼠肺组织样本;联合运用超滤辅助蛋白质样本制备技术、磷酸化肽段富集技术、TMT体外标记定量策略、反向预分级技术及高精度质谱技术进行肺腺鳞癌转分化过程中蛋白质组和磷酸化蛋白质组的时序性图谱的初步绘制,这一数据集包括了精确定量的9706种小鼠肺组织蛋白质和14807个小鼠肺组织蛋白质磷酸化位点。
A well-characterized mouse model recapitulating the malignant transformation progression from normal pulmonary epithelial cells to lung adenocarcinoma and then to lung squamous cell carcinoma was used to generate temporal proteomic and phosphoproteomic datasets for critical stages of lung adenosquamous carcinoma progression. Task 1 provided Task 2 with 27 lung tissue samples harvested from 17 mice at multiple time points of the lung adenosquamous carcinoma transdifferentiation mouse model (genotype: KrasG12D/Lkb1-/-). Task 2 selected a total of 15 lung tissue samples from 5 critical time points, with each time point having no less than 3 biological replicates. Combined approaches including filter-aided sample preparation (FASP), phosphopeptide enrichment, TMT in vitro labeling-based quantitative strategy, reverse-phase pre-fractionation, and high-resolution mass spectrometry were employed to preliminarily construct the temporal proteomic and phosphoproteomic atlases during lung adenosquamous carcinoma transdifferentiation. This dataset contains precisely quantified 9706 mouse lung tissue proteins and 14,807 phosphorylation sites of mouse lung tissue proteins.
