N6-methyladenosine (m6A) is the most abundant mRNA modification in mammalian cells, mediated co-transcriptionally by a methyltransferase ‘writer’ complex containing METTL3 as the S-adenosyl methionine (SAM)-binding subunit as well as adaptors such WTAP and ZC3H13 To profile m6A mRNA modification sites transcriptome-wide with single nucleotide resolution, we performed enhanced crosslinking and immunoprecipitation (m6A-eCLIP) in WTAP-, ZC3H13-, METTL3-deficient and NTC control cells derived from two healthy donors.
