Heat stress disrupts the function of dairy cow mammary epithelial cells and the m6A demethylase FTO is a key regulator of epitranscriptomic modification To investigate the role of FTO in the heat stress response of bovine mammary epithelial cells MAC-T we designed a 2x2 factorial experiment 1 37 normal temperature FTO overexpression plasmid transfection 2 37 empty vector transfection control 3 42 heat stress FTO overexpression plasmid transfection 4 42 empty vector transfection control RNA sequencing was performed to compare transcriptomic differences among the four groups This project aims to 1 identify FTO mediated differentially expressed genes under normal and heat stress conditions 2 reveal the regulatory mechanism of FTO in MAC-T cell thermal adaptation 3 provide transcriptomic data for clarifying the epitranscriptomic role of FTO in dairy cow heat stress response The generated data will support research on mammalian thermal biology and FTO related molecular breeding strategies for dairy cows
