M6A-seq & RNA-seq analysis of wild type and Mettl3-cKO liver tissues' or hepatocytes' transcriptomes

To investigate the role of METTL3-mediated m6A modification in liver, we performed m6A-sequencing and RNA-seq to map the m6A modification and gene expression in liver tissues or hepatocytes of wild type (WT) and liver-specific Mettl3-KO mice. m6A-seq and transcriptome profiles of liver in 8 weeks wild type (WT) and liver-specific Mettl3-KO mice were generated by deep sequencing,with biologic replicates. In addition, m6A-seq and transcriptome profiles of hepatocytes in 8 weeks wild type (WT) and liver-specific Mettl3-KO mice were generated by deep sequencing too.

为探究METTL3介导的N6-甲基腺苷(m6A)修饰在肝脏中的功能,我们通过m6A测序(m6A-sequencing)与RNA测序(RNA-seq)技术,对野生型(wild type, WT)及肝脏特异性Mettl3基因敲除小鼠的肝脏组织或肝细胞中的m6A修饰与基因表达水平进行了图谱绘制。 本研究通过深度测序结合生物学重复,获取了8周龄野生型与肝脏特异性Mettl3敲除小鼠的肝脏m6A测序及转录组测序数据。 此外,本研究还通过深度测序,获取了8周龄野生型与肝脏特异性Mettl3敲除小鼠肝细胞的m6A测序及转录组测序数据。
