Development of mitochondrial DNA cytochrome c oxidase subunit I primer sets to construct DNA barcoding library using next-generation sequencing

The project goal is to develop new mitochondrial DNA cytochrome c oxidase subunit I (mtDNA COI) primer sets which are designed to amplify the region efficiently using next-generation sequencing (NGS) system. Although the mtDNA COI primer is the most traditional barcoding primer set, primers targeting this region for NGS-based analysis had not been developed until now. In this project, we confirmed the universality of the primer sets which we designed using some insect species taxa. As a result, the developed primer sets were found to be suitable for 5 insect taxa (i.e., Coleoptera, Lepidoptera, Diptera, Ephemeroptera, and Trichoptera). Thus, the developed primer sets will contribute to enhance DNA barcoding library.
