CyclinD2-mediated regulation of neurogenic output from the retinal ciliary margin is perturbed in albinism

Ciliary margin zone cells have differential neurogenic potential in albino and pigmented eyes. We used single cell RNA sequencing (scRNA-seq) of albino and pigmented eyes to determine transcriptomic changes underlying differential RGC genesis. Peripheral retina cells containing ciliary margin zone was isloated using a-Cre;Td mice on albino and pigmented backgrounds. Cells were isolated by Fluorescence-activated cell sorting (FACS) according to the presence TdTomato signal and analyzed using scRNAseq.
睫状缘区(Ciliary margin zone)细胞在白化眼与色素性眼中具有差异化的神经发生潜能。我们通过对白化及色素性眼部样本开展单细胞RNA测序(single cell RNA sequencing, scRNA-seq),旨在明确介导视网膜神经节细胞(Retinal Ganglion Cell, RGC)差异化发生的转录组学变化。我们在白化与色素性遗传背景下,利用a-Cre;Td小鼠分离得到包含睫状缘区的外周视网膜细胞。随后根据TdTomato荧光信号的表达情况,通过荧光激活细胞分选(Fluorescence-activated cell sorting, FACS)分离目标细胞,并借助scRNA-seq完成后续分析。
