Methadone induces phospholipidosis and lysosomal dysfunction in astrocytes in vitro and in the developing brain

This dataset was generated to investigate the effects of subchronic methadone exposure on the lipid profile of primary neonatal rat astrocytes. As methadone is a standard treatment for opioid dependence during pregnancy, this study aimed to understand its effects on lipid homeostasis in developing brain cells. Primary rat astrocytes were cultured and treated with either 10 µM methadone or a vehicle control for three days. Hydrophobic (lipid) extracts were analyzed by untargeted lipidomic profiling. The analytical platform consisted of a Bruker Elute UHPLC system (using a C18 column) coupled to a Bruker compact Q-TOF mass spectrometer. Data was acquired in both positive ionization mode using an Auto-MS/MS (DDA) method.
