Transcriptomic analysis of iPSC tri-culture (microglia, neurons, astrocytes) stimulated with Tnf, Smac, and Zvad (TSZ) to trigger necroptosis pathway activation, in the presence or absence of a Ripk1 inhibitor Overall design: Cells were pre-treated for 1 hour with 1 μM Smac mimetic LCL161 (Selleck Chem, S7009), 20 μM zVAD-fmk (Enzo Life Sciences, ALX-260-020), and 1 μM RIPK1 inhibitor GSK’547 prior to a 4 hour incubation with 50 ng/mL recombinant human TNF (R&D Systems, 210-TA). Four hours after treatment, cells were collected and sequenced using the 10x Genomics platform.
