Cell-based microbiota analyses of a mock bacterial community and murine cecal content.

To evaluate the efficacy of our newly-developed cell-based quantitative method, Barcoding Bacteria for Identification and Quantification (BarBIQ), we measured a mock community containing 10 cultured human gut bacterial strains by BarBIQ. To analyze murine intestinal microbiota at single cell level with high taxonomic resolution, we surveyed the microbiota at two cecal locations from three co-housing C57BL6/J male mice by BarBIQ. For these cecal samples, we counted > 10^4 cells per sample, determined totally more than 600 cOTUs, and identified more than 700 16S rRNA sequences with single base accuracy.
