Assessing global transcriptomic changes in signaling using RNA-seq. Overall design: NCI-H358 cells were seeded as 1,000 cell spheroids in Aggrewells and allowed to equilibrate for 48h. Afterwards, cells were treated for either 6h (initial signaling) or 72h (rebound signaling). The respective samples, were treated with different compounds, either as monotherapy or in combination using a KRAS G12C inhibitor (MRTX849) or a SOS1::KRAS inhibitor (BI00923406).
