Comparison of proteome between Candida tropicalis U1 with uracil deficiency in shaking and stand culture

Activated tropical Candida was inoculated into 50mL of YPD liquid medium at a 1% inoculation rate. Two groups were set up, with three replicates in each group. One group was placed in a 30 ℃ constant temperature incubator for static cultivation , while the other group was placed in a shaking table at 30 ℃ and 180 r/min for shaking cultivation (S). Select bacterial cultures that have been cultured for 8 hours in the stationary phase, 14 hours in the logarithmic growth phase, and 28 hours in the stable phase. Wash the cultures three times with PBS buffer, add 5 mL of buffer to resuspend the bacterial cells in an EP tube, centrifuge to remove the supernatant, rapidly freeze with nitrogen for 10 minutes, and store in a -80 ℃ freezer before sending them to Meiji Biotech. Using iTRAQ (Isobar Tags for Relative and Absolute Quantification) technology to achieve qualitative and quantitative analysis of proteins by labeling protein or peptide ends.
