资源简介:
Entry is a part of Mechanomics collection (10.6084/m9.figshare.c.5399826) that contains materials supporting findings presented in the following manuscript: Urbanska, M., Ge, Y. et al., De novo identification of universal cell mechanics gene signatures BiorXiv: https://doi.org/10.1101/2021.04.26.441418; first published: 27th April 2021, last updated: 4th Nov 2024 eLife: https://doi.org/10.7554/eLife.87930; first published: 23rd March 2023, last updated: 27th Dec 2024 This dataset contains mechanical measurements of murine induced pluripotent stem cells (iPSCs) of two different classes: • transgene-dependent, fuzzy-colony forming - F-class • transgene-independant, compact-colony forming - C-class. These two cell types were first characterised by Tonge et al (Nature, 2014). The cell lines used in this study were derived in a previously published study by Urbanska et al (Development, 2017) via reprogramming of neural progenitor cells using a dox-induced expression of the OSKM reprogramming factors. Measurements were performed using real-time deformability cytometry (RT-DC). _rtdc.zip - contains data acquired during the measurments in '*.rtdc' format _tsv.zip - contains extended data in a '*.tsv' format _glmm.zip - contains result summaries of statistical analysis comparing Young's moduli of the different cell types using generalised linear mixed effects models Tonge, P. D. et al. Divergent reprogramming routes lead to alternative stem-cell states. Nature 516, 192–197 (2014) https://doi.org/10.1038/nature14047 Urbanska, M. et al. Single-cell mechanical phenotype is an intrinsic marker of reprogramming and differentiation along the mouse neural lineage. Development 144, 4313–4321 (2017) https://doi.org/10.1242/dev.155218
