Gene expression of donor monocytes recovered from various tissues of recipient mice one day after transfer

Monocyte are recruited to almost all peripheral tissues. Micro-environmental stimuli appeared to alter monocyte transcription once they enter the intestine and their subsequent development in the tissue. We therefore investigated whether there is tissue specific monocyte adaption and differentiation in vivo by examining donor monocytes in various tissues i.e. small intestine, liver, spleen and BM. Transferred monocytes recovered and FACS-sorted from the small intenstine, liver, spleen and BM of recipient mice at different time points and used for gene expression analysis. Following sorting, cDNA was made immediately and preamplified with Multiplexed target enrichment (MTE) primers. Hybridization of pre-amplified DNA with target specific oligonucleotide probe pairs were performed using a nanostring technology.
