Comparative analysis of four murine Treg subsets differentially affected by pre-transplant conditioning and acute gastrointestinal GvHD following allogeneic hematopoietic stem cell transplantation. Four different Treg subsets were FACS sorted and subjected to comparative transcriptome profiling in a pairwise manner, using the following gating stategy: Ly/singlet/Live/CD4+/Foxp3+/Nrp1+/OX40bright Treg cells were sorted from six act-mOVA/Foxp3GFP mice, one sample each, and compared to Ly/singlet/Live/CD4+/Foxp3+/Nrp1-/OX40dim Treg of the same animals, one sample each (animals#1-6), serving as reference. Next, Ly/singlet/Live/CD4+/Foxp3+/Nrp1+/ITGB7+ Treg were sorted from six independent OVA/Foxp3GFP mice (animals#7-12), and compared to Ly/singlet/Live/CD4+//Foxp3+/Nrp1-/ITGB7- Treg of the same animals .
