To investigate the effect of oncogenic mutations on genome-wide DNA methylation in stem cells, we performed an Infinium MethylationEPIC BeadChip assays. To clarify that the epigenetic change is caused only by one oncogenic mutation, we introduced each oncogene into individual wt hESCs, using dox to overexpress the GoF mutation gene for nine days. The DNA methylation profiles demonstrated that the seven mutant hESCs and wt hESCs as control (empty vector) were distinguished by DNA methylated gene loci. We introduced each oncogene into individual wt hESCs, using dox to overexpress the GoF mutation gene for nine days, then performed an Infinium MethylationEPIC BeadChip assays. Stanford Functional Genomics Facility (SFGF) using the Infinium MethylationEPIC array which interrogates over 850,000 methylation sites at single-nucleotide resolution.
