This study utilized a fully MHC-mismatched mouse model of heart transplantation (Balb/c donors to C57BL/6 recipients) to investigate the immunomodulatory mechanism of Icaritin (ICT) . Recipient mice were treated with ICT for 7 days post-transplantation. Splenocytes were then harvested for single-cell RNA sequencing (10x Genomics platform). The data aims to delineate the comprehensive landscape of immune cell subsets and their transcriptional changes within the spleen under allogeneic rejection and ICT treatment, with a particular focus on T cell differentiation and activation pathways. This dataset provides a resource for understanding how ICT attenuates rejection and may inform its application in transplant immunotherapy.
