Uncovering genetic dependencies in the DNA repair network after ionizing radiation

We constructed a combinatorial CRISPR guide library to target ~200,000 pairwise combinations of 461 genes associated with DNA Repair. Overall design: eHAP-iCas9 cells harboring this library were grown under standard growth conditions as well as an LD30 dose of ionizing radiation to induce global DNA damage. Each condition grew for 14-15 days before being collected and sequenced.
