We previously demonstrated that overexpression of FBXL10, a demethylase for histone H3K36, induced leukemia in mice (Ueda T. et al., Blood, 125: 3437-3446, 2015). To analyze the effect of FBXL10 deficiency on leukemogenesis, we generated mice lacking the long form of FBXL10 that contains the demethylase domain (Fbxl10-/- mice). We found that lack of the catalytic domain of FBXL10 exhibited a significant delay in MLL-AF9-mediated leukemia in a mouse BMT model.
