Title: Transcriptome analysis of Triptolide regulated M2 macrophage polarization in asthma researchDescription:Asthma is a chronic inflammatory airway disease characterized by airway hyperresponsiveness, mucus overproduction, and airway remodeling. Macrophages play critical roles in the initiation and resolution of airway inflammation. In particular, alternatively activated M2 macrophages, induced by cytokines such as interleukin-4 (IL-4), are known to contribute to tissue repair, airway remodeling, and immune regulation. Understanding how M2 polarization is regulated may provide novel insights into the mechanisms of asthma pathogenesis and potential therapeutic strategies.Triptolide (TRP), a diterpenoid triepoxide extracted from Tripterygium wilfordii Hook F, has well documented anti-inflammatory and immunomodulatory properties. However, its regulatory effects on macrophage polarization and the underlying molecular mechanisms remain incompletely understood.In this study, we performed transcriptome profiling using the murine macrophage like RAW 264.7 cell line to investigate the effect of TRP on IL-4 induced M2 polarization. Two experimental groups were analyzed: (1) RAW 264.7 cells polarized with IL-4 into M2 macrophages, and (2) IL-4 treated RAW 264.7 cells further exposed to TRP. Six biological replicates were included per condition to ensure robust analysis. RNA sequencing (RNA-seq) was conducted to characterize global gene expression changes associated with TRP treatment in the context of M2 polarization.The dataset provides valuable resources for dissecting the molecular mechanisms by which TRP regulates macrophage polarization. These findings may shed light on the immunomodulatory role of TRP in asthma and contribute to the development of novel therapeutic approaches targeting macrophage mediated airway inflammation.
