Treg cell-specific ablation of Cxxc1 leads to the development of a lethal inflammatory disease characterized by spontaneous T-cell activation.Cxxc1-deficient Treg cells showed reduced immunosuppressive capacity without affecting Treg cell numbers.To investigate the role of Cxxc1 as a transcriptional regulator of Treg subsets, we performed single-cell RNA-seq (scRNA-seq) paired with T cell receptor sequencing (TCR-seq) . CD4+YFP+ Treg cells from WT and Foxp3Cre Cxxc1fl/fl mice were isolated for scRNA-seq.
调节性T细胞(Treg cell)特异性敲除Cxxc1可诱发致死性炎症疾病,该疾病以自发性T细胞活化为特征。Cxxc1缺陷的调节性T细胞免疫抑制能力降低,但不影响调节性T细胞的数量。为探究Cxxc1作为调节性T细胞亚群转录调控因子的作用,我们开展了单细胞RNA测序(single-cell RNA-seq,scRNA-seq)与T细胞受体测序(T cell receptor sequencing,TCR-seq)的配对分析。我们分离了野生型(WT)小鼠及Foxp3Cre Cxxc1fl/fl小鼠的CD4+YFP+调节性T细胞用于单细胞RNA测序。
