The SPT-2 histone chaperone is involved in the maintenance of chromatin structure during transcription. Here, we performed mRNA-seq to compare gene expression profiles of spt-2 mutant worms. We compare two full spt-2 gene knock-out worm strains (KO-A and KO-B), or a strain expressing an spt-2 allele with reduced binding to histones (M627A), to wild type N2 worms. We performed mRNA-seq comparing RNA extracted from spt-2 mutant worms to WT control; worms were collected as synchronised adults. The experiment was performed in triplicate. We first compared WT to spt-2 KO worms (Batch 1) and then WT to spt-2 M627A worms (Batch 2).
