hMSCs and human glioma cells U251 were purchased from Cyagen Biosciences Inc. (Guangzhou, People’s Republic of China), and human glioma cells U87 were obtained from the American Type Culture Collection (ATCC). Those cells were cultured in DMEM supplemented with 10% FBS, and incubated at 37℃ with 5% CO2. To co-cultured hMSCs and human glioma cells in vitro, a system with 0.4 µm pore cell culture chambers embedded into 6-well plates was applied. hMSCs passaged to the fifth generation were adjusted to 105/L and 2 mL of cells were planted into the 6-well plate and cultured for 24 hours, and then the cell culture chamber with 0.4 µm pore were inserted into the 6-well plate. 2 mL of human glioma cells U251or U87 with the cell concentration of 105/L were added to the upper chamber. In the control group, 2 mL of cell culture medium alone were added into the upper chamber. The medium was changed every day for 7 days. The morphology of cells in each group was observed by inverted microscope.
