We report global transcriptional profiles of epidermal basal cells from plantar skin of control mice, high fat diet-fed (HFD) mice and aged mice. By RNA-sequencing of FACS-isolated interfollicular epidermal (IFE) basal cells, we revealed that calcium signal-associated genes, which regulate differentiatin of keratinocytes,were enriched in the plantar skin of HFD and aged mice. Overall design: For RNAseq, mRNA profiles of FACS-isolated interfollicular basal cells (CD49f+, CD34-) from plantar skin of control (nomal diet fed) mice, high fat diet fed (HFD) mice and aged mice were generated by deep sequencing, in triplicate, using Illumina NextSeq500. For scRNAseq, after dissociation of the dermis into single cells, we generated chromium single cell 3' v2 libraries from randomly captured dermal cells and sequenced them using Illumina NextSeq500. We profiled 13,584 cells (Cont: 4,775 cells; HFD: 4,184 cells; Aged: 4,625 cells) with a range of 20,000–40,000 mean reads per cell for each sample, whereby approximately 1,400–1,600 median genes per cell for each sample were detected.
