In order to search for pharmacodynamic biomarkers of a TREM2 agonist antibody (Para09) in primates we performed bulk RNA-Seq on cortical tissue from cynomolgus monkeys after single IV injected doses. Changes in proliferation genes such as MKI67 were observed, as well as genes associated with glial activation, such as CHI3L1. This experiment demonstrates that our antibody induces transcriptional changes in the brain after peripheral administration. Overall design: Monkeys were treated with anti-hTREM2 agonist Para09 or an isotype control via intrathecal injection and brain samples were collected 2 days later. Bulk RNA was extracted from frontal cortex. Please note that the treatment 1, 2, 3 represents: treatment 1: "Anti-gD 200 mg/kg" treatment 4: "Para09 50 mg/kg" treatment 5: "Para09 200 mg/kg"
