This project provides a streamlined experimental workflow for cell-free transcriptomics, in two key steps: (i) optimizing the cell-free buffer using active learning and (ii) directly sequencing long-read RNA from a cell-free reaction. All transcriptomics data were generated using long-read RNA-direct sequencing on a MinION by Oxford Nanopore Technologies. The dataset includes: in vivo transcriptome of T7 phage in E. coli MG1655, 10 minutes post-infection, in vitro transcriptome of the T7 genome expressed in cell-free with the "mRNA-optimized" buffer, in vitro transcriptome of the T7 genome expressed in cell-free with the "trade-off transcription-translation" buffer, in vitro transcriptome of the T7 genome expressed by purified T7 RNA polymerase, in vitro transcriptome of the T7 genome expressed by purified E. coli RNA polymerase (sigma 70)
