This deposition contains the data files and R code generated in the course of our study entitled "Flow Cytometry Analysis of Perturbations in the Bacterial Cell Envelope Enabled by Monitoring Generalized Polarization of Solvatochromic Peptide UNR‑1" published in Analytical Chemistry 2025 (DOI: 10.1021/acs.analchem.4c04953).
Our publication described a new way of studying the state of the bacterial cell envelope using ratiometric flow cytometry. In brief, bacteria were stained with the environmentally sensitive fluorescent probe UNR-1 and analyzed by flow cytometry. The value of generalized polarization (GP) of fluorescence was calculated for each detected cell using the R programming language. GP was derived from the fluorescence intensities of UNR-1-labeled cells detected in the yellow and red emission channels. GP of UNR-1 was a convenient parameter for estimating the state of bacteria. For instance, dramatic drops of GP were observed for cells exposed to environmental stress (high temperatures, antibiotics, etc.).
The deposition includes examples of raw data files, processed data files, and the R code used to analyze the flow cytometry data and generate figures used in our publication in Analytical Chemistry.
