Transcriptome analysis of Arabidopsis wild-type and kam2 seedlings grown on sucrose-free medium

We performed RNA-seq analysis to investigate why the kam2 mutant undergoes seedling arrest on sucrose-free medium. We used Arabidopsis wild-type and kam2 seedlings grown on solid medium (0.2 or 0.5% gellan gum) or solid medium (0.5% gellan gum) covered with plastic sheets, all on sucrose-free medium at 4 days after sowing. Total RNA was extracted from a pool of 20 wild-type and kam2-2 seedlings per condition, and five biological replicates were performed (30 samples in total). RNA-seq was performed using a newly developed library preparation method called Lasy-seq.
