Fourteen healthy female adults (aged between 22 and 35 years old) and ten female elderlies residing in a nursing home (aged between 70 and 94 years old) were recruited for this study. All participants signed an informed declaration consent and answered a structured questionnaire to collect anthropometric (age, weight, height and body mass index) and clinical data. This study was approved by the Ethics Committee of the Faculty of Medicine of the University of Coimbra (reference CE-144/2016). Personal data were treated in compliance with current European data protection rules [Regulation (EU) 2016/679 and law No 58/2019]. For mitostress and glycostress analysis UDSCs were seeded at a density of 20,000 cells/80 μL/well in a 96-well Seahorse XF96 V3 PS cell culture microplate. For ATP analysis, 20,000 cells/80 μL/well (60,000 cells/cm2) were plated on a white opaque-bottom 96-well plate.
本研究招募14名健康成年女性(年龄22至35岁)及10名居住于养老院的老年女性(年龄70至94岁)参与。所有参与者均签署知情同意声明,并填写结构化问卷以收集人体测量学数据(年龄、体重、身高及身体质量指数(Body Mass Index))与临床资料。本研究已通过科英布拉大学医学院伦理委员会批准(批准编号:CE-144/2016)。个人数据的处理严格符合现行欧洲数据保护规范[欧盟条例(EU)2016/679及第58/2019号法律]。为开展线粒体应激(mitostress)与糖应激(glycostress)分析,将UDSCs(尿源干细胞)以20000个细胞/80μL/孔的密度接种于96孔Seahorse XF96 V3 PS细胞培养微孔板中。针对三磷酸腺苷(ATP,Adenosine Triphosphate)分析,以20000个细胞/80μL/孔(60000个细胞/cm²)的密度将细胞铺板于白色不透明底部96孔板内。
