TRIM21 catalyzes K63-linked ubiquitination of PD-1, stabilizing it by counteracting K48-linked ubiquitination and degradation. TRIM21 knockout reduces PD-1 levels, activates CD8+ T cells, enhances tumor sensitivity to anti-CTLA-4 therapy, and improves the anti-tumor efficacy of anti-CD19 CAR T cells.
