The wound epidermis is required for limb regeneration. To elucidate the roles of the wound epidermis during early regeneration, we examined how the transcriptional programs of early dividing cells (enriched for blastemal progenitors) and non-dividing cells in regenerating stump tissues as well as epithelial cells change when we prevented wound epidermis formation. To prevent wound epidermis formation, we performed full skin flap surgeries to inhibit epithelial migration over the exposed stump tissues post-amputation. We separately sequenced stump-derived 4N, stump-derived 2N, and epithelial cells from the sutured full skin flap at 5 days post-amputation (dpa). We sequenced a total of 9 samples, 3 biological replicates per timepoint. These samples were multiplexed and sequenced on 6 lanes on a Hiseq 2500. (dataset from Tsai et al.
