A separate dormancy simulation experiment was conducted at the same facility using Pinus tabuliformis seeds from Pingquan City, Hebei Province as follows: 1. Post-germination, 2000 uniformly growing seedlings were transplanted into 6 cm pots, with 2-4 seedlings per pot; 2. Seedlings were grown in a greenhouse under the same conditions as described earlier for three months; 3. To induce dormancy, 1800 seedlings were transferred to an SD-W climate chamber for 8 weeks to promote bud-set formation; 4. After bud-set rates exceeded 90%, the experimental group was moved to SD-C for 7 weeks to simulate winter and transition to endodormancy; and 5. The SD-Cgroup was then moved to LD-W conditions for two weeks to induce dormancy release. Sampling was conducted at five time points: week 0, 1, 3, and 7 of phase 4, and week 2 of phase 5. Each sampling included three biological replicates of shoot apices for transcriptome sequencing (minimum 200 mg per biological replicate), with each replicate consisting of pooled apical shoot samples from 10-30 seedlings.
