Single Cell sequencing analysis of Bone Marrow samples

Study of hematopoietic stem and progenitor cell (HSPC) hierarchies in bone marrow cells from myelodysplastic syndromes through the transcriptional analysis of single cells in distinct differentiation states Lin-CD34+ HSPCs were FACS-purified from magnetically CD34-enriched mononuclear cells isolated by density gradient from bone marrow aspirates. A total of 3000 cells were sorted and input onto the Chromium Single Cell A Chip Kit (10x Genomics, Pleasanton, CA), in which single cells were lysed and barcoded for reverse-transcription. The pooled single-stranded, barcoded cDNA was amplified and fragmented for library preparation.
