Cell wall damage causes the induction of plant signaling. Here, we used isoxaben treatment to inhibit cellulose biosynthesis and cause cell wall damage. PEP3 is detected by PEPR receptors and contributes to both plant defense signaling and the regulation of isoxaben-induced responses. Overall design: Arabidopsis thaliana Col-0 seedlings (10 days-old) were treated with DMSO (mock control), 600 nM isoxaben (ISX), 10 nM PEP3 and a combination of ISX and PEP3. Samples were taken at 6 h after treatment and 3 biological replicates were analyzed per treatment.
