To observe the tumor promotion effect of diabetes,8-week-old male C57BL/6 mice were fed high-fat diet (Research Diet,D12492,60% fat) for 4 weeks to induce insulin resistance. Streptozocin (STZ, Sigma-Aldrich, S0130) at a dose of 40mg per kg body weight was injected intraperitoneally for 5 consecutive days after 4-week HFD feeding. 10 Days after the last dose of STZ,serum glucose was measured after a 10-hour fast, mice with glucose level over 11.1mmol per L were selected for experiments. For the wild-type control group(WT), C57BL/6 mice were fed normal diet for 4 weeks followed by citrate buffer injected intraperitoneally. 1*106 Pan02 cells were injected into the pancreas capsule of WT and DM mice. Tumor-bearing mice were sacrificed 14 days after tumor inoculation.
