We used microarray technology to investigate the transcriptional consequences of loss of Gli3 from the anterior mesenchyme of the developing mouse limb bud at both E11.5 and E12.5. Comparisons were made between RNA derived from the anterior margin of Gli3Xt/Xt limb buds and that derived from wild type littermates Keywords: genetic modification The experiment contains a series of pairwise comparisons between staged mutant Gli3(Xt/Xt) and WT anterior portions of limb bud at 11.5dpc and 12.5dpc. Forelimbs and hindlimbs were studied independently given the developmental delay between limb buds.
本研究采用基因芯片技术(microarray technology),探究了发育中小鼠肢芽前间充质内Gli3基因缺失在胚胎发育第11.5天(E11.5)与第12.5天(E12.5)时所引发的转录组变化。实验以Gli3Xt/Xt突变纯合肢芽前缘提取的RNA,与野生型同窝仔鼠对应样本的RNA开展对比分析。关键词:基因修饰。本实验包含一系列配对比较实验,比对对象为受精后11.5天(11.5dpc)与12.5天(12.5dpc)的分期样本,即突变型Gli3(Xt/Xt)与野生型(Wild Type,WT)肢芽前缘组织。鉴于前后肢存在发育时序差异,本研究分别对前肢与后肢进行独立分析。
