Paraescarpia echinospica and Sclerolinum tubeworms were collected from the Haima Cold seep in the South China Sea. Paraescarpia echinospica was sampled in April 2023 at latitude and longitude (110.27E, 16.435N). Sclerolinum was sampled in July 2022 using the ROV robot. Paraescarpia echinospica was dissected with a sterile scalpel and scissors and sampled from the trophosome site. In contrast, Sclerolinum tubes were too thin and soft to be separated from the tissue, so the entire Sclerolinum was sampled directly. The samples were rinsed with DNA extraction buffer (100 mM Tris HCl, 100 mM EDTA, 100 mM Na2HPO4, 1.5 M NaCl, 1% CTAB, pH 8.0) to eliminate any remaining surface impurities and preservatives. Subsequently, the tissue blocks were placed in 1 mL of DNA extraction buffer, which was adequately cut and left to stand with sterile scissors. The upper suspension containing microbial cells was collected after the large pieces of tissue were precipitated. The microorganisms were ultimately precipitated through high-speed centrifugation. The resulting microbial precipitates were obtained for macrogenetics studies. The DNA was extracted and purified using a magnetic bead-based soil and fecal genomic DNA extraction kit (TIANGEN) according to the manufacturer's protocol. Qualitative and quantitative analysis was performed on the purified DNA using a Qubit 4.0 device and agarose gel electrophoresis with 1% agarose. The VAHTS Universal Plus DNA Library Prep Kit for Illumina was used for metagenomic library construction according to the manufacturer's protocol.Genome assembly was performed on the symbionts of Paraescarpia echinospica (Hai6C03) and Sclerolinum (SCTW1H) collected from Cold Seep, South China Sea. The Hai6C03 assembled genome had a size of 4,171,724 bp, a GC content of 54.43%, a completeness degree of 99.33%, and a contamination degree of 1.6%. The genome assembled by SCTW1H was 2,832,488 bp in size with a GC content of 50.46%. Its completeness was 94.72%, and the contamination degree was 0.23%.
