Nematodes have been known as major factors in crop loss and important players in nutrient cycling. For enhancing productivity of agricultural fields, better understanding of their environments may be of essential importance. The three primer sets reviewed in this study worked well for NGS metabarcoding specifically for nematode species with a wide range of detection coverage. Among these primer sets, F548_A/R1912 was highly specific to nematode species and applicable to soil DNA. It is also necessary to be noted that another primer set, F548_A/R915 or F548_A/SSU26R using DNA from nematode suspension may extend the coverage to detect nematode species more than that using only F548_A/R1912 with soil DNA.
