The purpose of this study was to determine the roles of the flanking regions of the FLI ETS DNA-binding domain in gene regulation of Ewing sarcoma cells. EWS/FLI cDNA constructs including various deletions of the FLI portion were virally transduced into cells and we sought to determine the transcriptional profile of each. Overall design: Endogenous EWS/FLI was knocked-down in the Ewing sarcoma A673 cell line and EWS/FLI cDNA constructs were expressed in these cells. 3 biological replicates of A673 cells containing the cDNA constructs were collected after 8 days and total RNA was extracted to study the transcriptomic profiles of each EWS/FLI construct. Data used for analysis and creation of Figure 4.
