Mouse Splenocyte Enrichment Strategies via Negative Selection for Broadened Single Cell Transcriptomics

Mammalian splenic tissue is rich in functional immune cells, the majority of which are lymphocytes. Evaluating populations of low abundance cells can therefore be problematic, and classic enrichment protocols may activate immune cells prior to evaluation via binding to cell-specific surface receptors. This protocol enriches minority immune cell populations from the mouse spleen via immunomagnetic negative depletion, thus permitting downstream analyses such as single-cell RNA sequencing (scRNA-seq) that otherwise may be affected by traditional methods of enrichment.
