Agrobacterium-transformed yeast BY4741 whole genome sequencing

To study potential mutagenic effects of Agrobacterium VirD5, haploid yeast BY4741 was transformed with Agrobacterium tumefaciens (LBA1100) wild type for virD5 or with a virD5 mutant. Of about 20,000 G418-resistant colonies (G418 to select for the presence of the T-DNA) obtained from the transformation by the wild type Agrobacterium, one was auxotrophic (strain LBYR50) and 9 were thermosensitive (of which one was named strain LBYR43). None of colonies transformed by the VirD5 mutant were auxotrophic or thermosensitive. The wild type strain (LBYR1) and mutants LBYR50 and LBYR43 were sequenced.
