This study used Siberian sturgeon as the experimental animal. We Cloned the CDS sequences of nmur1 and nmur2 and detected their organizational distribution patterns. Then, we clarified their response patterns to different nutritional states of the body, and their roles in the appetite suppression process of NMU-25. In pre-prandial experiment, Siberian sturgeons (n=84) were randomly dispersed in 7 groups. Five of the treatments were fasted at a specific time point (14:00), while the other two treatments fed at the feeding time point (14:00). Randomly collect 9 fish from each treatment, including five fasted treatments at -3 h (11:00), -1 h (13:00), 0 h (14:00), +1 h (15:00) and +3 h (17:00) and the control of above treatments that is two fed treatments at +1 h (15:00) and +3 h (17:00). hypothalamus, valve intestine, and duodenum were collected.In fasting and refeeding experiment, Siberian sturgeons (n=180) were randomly dispersed in 15 group. 6 unfed groups were sampled on different fasting days 1, 3, 6, 10, 15, and 17day, respectively. Simultaneously, 6 fed groups were sampled on 1, 3, 6, 10, 15, and 17day, which served as control for respective days. The remaining 3 groups were refed daily between days 10to 17 of fasting and sampled on 10, 15, and 17 day (re-fed for 1, 5, and 7 days). hypothalamus and liver were sampled.In long-term i.p. and i.c.v. injection experiment, Siberian sturgeons (n=48) were randomly divided into 4 groups. The first group received long-term i.p. injection with NMU-25 (100 ng/g BW), and the second group was injected with saline for 7 days as control. Similarly, the third group received once i.c.v. injection with NMU-25 (10 pmol/g BW ), and the last group was injected with saline as control. Hypothalamus and valve intestine were sampled.The genes tested in this study are nmur1 and nmur2. The qPCR instrument brand used is Bio rad.
