Purpose: We investigated the function of B cells in the liver and spleen of mice hepatitis model by RNA-seq analysis. Method: To induce the hepatitis, adeno-associated virus 8 (AAV8) which mediates overexpression of IL-12 selectively in the liver was administered to C57BL/6J mice. AAV8 mediating overexpression of luciferase was administered to control mice. We isolated B cells from liver and spleen of these mice at week 4. Gene expression profiling analysis was performed using data obtained from RNA-seq of 4 different group of B cells (B cells in the liver of control mice; B cells in the liver of hepatitis mice; B cells in the spleen of control mice; B cells in the spleen of hepatitis mice). 3 samples were obtained for each group (n=3). B cells were isolated as CD45+ CD3- CD19+ CD138- cells by FACS-sorting.
