We performed ChIP-Seq on MV4;11 WT, CDK12AS/NULL, CDK13AS/AS, #1CDK12AS/NULL;CDK13AS/NULL and #2CDK12AS/NULL;CDK13AS/AS MV4;11 clones treated with 1-NM-PP1 or vehicle for 4 hours to determine the location of total Pol II and phosphorylated Ser2 of Pol II occupancy Overall design: Analog senstive MV4;11 cells were treated with 1-NM-PP1/DMSO for 4 hours, then total Pol II or phosphorylated Pol II at Ser2 of the CTD bound chromatin were isolated from soniacated nuclear extracts using antibody angaist RNA polymerase II, clone CTD4H8 mouse monoclonal or RNA polymerase II (phospho CTD Ser-2) respectively
