We report the single cell transcriptome of CD45+ cells from the livers of bone marrow chimeric wildtype mice with NASH that had previously been transplanted with bone marrow from TREM2-deficient or wildtype littermate mice. We show that hematopoietic TREM2 deficiency is associated with alterations in the hepatic immune cell composition in the context of NASH. Overall design: Male C57Bl6/J mice were lethally irradiated and transplanted with bone marrow from Trem2-/- or Trem2+/+ littermates. Following 6 weeks recovery, mice were placed on a methionine/choline-deficient (MCD) diet for 4 weeks to induce non-alcoholic steatohepatitis (NASH). Live (7AAD-) CD45+ cells were sorted from the livers of these mice and pooled in equal proportions from 4 mice per group, which were subsequently subjected to 5' single cell sequencing.
