The goal of this study is to examine age-related changes in the beta-1 adrenergic receptor dependent vasodilaiton in coronary microvessels from young, old, and old rats treated intravenously with stromal cells isolated from adipose tissue (SVF). We have shown that an aged coronary microvasculature has impaired vasodilation to adrenergic agonists and that a single treatment of SVF cells ameliorates the impaired vasodilation but the therapeutic mechanism is unknown. Overall design: Female Fischer-344 rats aged 3 months (young) 24 months (old), and old injected with a single intravenous injection of 10^7 adipose-derived stromal vascular fraction cells four weeks prior to sacrifice were used. Coronary arterioles less than 150 um were isolated for isolated vessel pressure myography experiments or stored at -80 degrees for RNA isolation. RNA was isolated from = 5 microvessels from pooled within the group using RNAeasy Micro plus kit (Qiagen). RNA sequencing will be performed and data analyzed for age-related and cell treatment related changes in the intracellular proteins of the beta-adrenergic vasodilatory signaling cascade as well as proteins for receptor modification, internalization, recycling, and proteins involved in oxidative stress.
