PCBP1 regulates transcription by binding ssDNA at promoters

To investigate the role of PCBP1 in transcription through its ability to bind DNA, we performed a CRISPR knockout of PCBP1 in A549 lung adenocarcinoma cells and generated a genome wide map of PCBP1's DNA binding using CUT&RUN. We used RNA-seq of A549 WT cells and two A549 PCBP1 KO cell lines we generated to quantify gene expression changes. We used PCBP1 CUT&RUN to generate a map of PCBP1's DNA binding sites. Overall design: Comparative gene expression profiling analysis of RNA-seq data for A549 cells and PCBP1 KO derivatives. Analysis of PCBP1 DNA binding using Cleavage Under Targets & Release Under Nuclease (CUT&RUN). Two biological replicates for PCBP1, IGG, and H3K4me3 (positive control) antibodies.
